Calibration Procedure
- Spot a sample of a the appropriate calibration mix and load an appropriate method
- Collect the spectra with the default calibration and open it in Data Explorer
- Baseline correct the spectrum (Process > Baseline Correction)
- Filter noise from the spectrum (Process > Noise Filter/Smooth)
- Manually calibrate the spectrum
- Select Process > Mass Calibration > Manual Calibration
- A window titled Manual Mass Calibration appears, move this window off to the side so you can see all of the peaks of interest
- Select the first peak to calibrate on by using the right mouse button to draw a box around the entire peak
- From the Select or Create Reference Peak Information select the correct mass for the peak you are calibrating and click OK
- Repeat for each calibrant peak
- After identifying all of your peaks click the Plot button in the Manual Mass Calibration window - your peaks will now reflect the new calibration settings
- Click Export to save your calibration settings as a .cal file
- Load the calibration file into the Voyager Instrument Control Software
- In the lower right hand corner of the window select External Calibration and browse to the .cal file you created above
- Collect your sample spectrum - the calibration settings will be automatically applied
Calibration Mixtures
Myoglobin/Cytochrome C
Mass Range: 6,000 to 17,000 Da
- Use a 1:10 dilution of 1 mg/ml Cytochrome C and 1 mg/ml Myoglobin in 50 H2O / 50 ACN + 0.1% TFA
- Mix 1:1 with appropriate matrix solution (recommendation: sinapinic acid)
Agilent Calibration Mix 1
Mass Range: 900 to 1,700 Da
Contains a mixture of Glu1-Fibrinopeptide B, Neurotensin, Angiotensin I, and des-Arg1-Bradykinin
- Use full strength
- Mix 1:1 with appropriate matrix solution (Recommendation: sinapinic acid - best, or α-cyano-4-hydroxycinnamic acid - OK)
Others
Any combination of the above calibrant mixes, or a solution of any synthetic peptide of known mass can be used to calibrate the maldi.
